Abstract Background: The research of anticancer drugs is still being developed, especially on the hydroxyurea group. Objective: This study aimed to compare the synthesis methods of 1-benzyl-3-benzoylurea derivatives, namely 1- benzyl-3-(4-ethyl-benzoyl)urea and 1-benzyl-3-(4-chloromethyl-benzoyl)urea. Methods: Optimization of the synthesis method was carried out by comparing the synthesized products with and without reflux heating. The products obtained were subjected to structure elucidation analysis by using FTIR, 1H-NMR, and 13C-NMR. Results: The reflux synthesis method produced by-product compounds, which needed to be separated by column chromatography. The method without heating is preferred because it did not produce by-product compounds. Conclusion: The method of synthesizing 1-benzyl-3-benzoylurea derivatives was recommended without using reflux heating, and in the future, researchers must find a better approach to get a better yield.
