Abstract: | Abstract Background: Kasturi (Mangifera casturi Kosterman) is one of the typical plants of Kalimantan which has potentially bioactive because it has a high phenol and flavonoid content. This plant is used in medicine by people for generations and is rarely found, so need to know the chemicals that are efficacious in the bark. Objective: This study aims to determine the total levels of phenolic and flavonoids of ethyl acetate fraction from the methanol extract of Kasturi bark. Methods: Qualitative analysis was done through phenolic phytochemical screening with FeCl3 reagent, whereas flavonoids using Mg-HCl, NaOH 10%, and H2SO4. Thin-layer chromatography (TLC) was also carried out with methanol eluent and 10% FeCl3 spotting appearance for phenolic compounds, 5% AlCl3 and citroborate for flavonoid compounds. Determination of total phenolic and flavonoid levels using colorimetric methods with the Folin-Ciocelteu complex reagents in phenolic and AlCl3 complex reactants on flavonoids whose absorbance was measured using UV-Vis spectrophotometer. Results: The results showed that the value of rendement of methanol extract of Kasturi bark is 10.38% and its ethyl acetate fraction is 4.2%. Phytochemical screening and TLC was identified phenolic and flavonoid content. The ethyl acetate fraction from the methanol extract of Kasturi bark contains a total phenolic level of 3.92 mg GAE/g fraction and a total flavonoid level of 5.14 mg QE/g fraction. Conclusion: The study can be concluded that the ethyl acetate fraction from the methanol extract of Kasturi bark contains flavonoid and phenolic compounds that can be used as alternatives natural medicine sources.
