Abstract: In ethnomedicine, Tetracera indica has many pharmacological properties, including the treatment of diabetes, hypercholesterolemia, fever, and gout. Since diseases caused by free radicals need to be counteracted with antioxidants, this research aimed to discover the active antioxidant extract using the FRAP method and toxicity test on 264.7 RAW cells. Extraction was performed by ultrasonic-assisted extraction with n-hexane, ethyl acetate, and 96% ethanol as solvents. Total phenol content was measured colourimetrically using Folin-Ciocalteu reagent and flavonoid content using AlCl3. Toxicity test with MTT on RAW 264.7 macrophage cells. The highest total phenol content was 210,229 mg GAE/g found in the 96% ethanol extract. The ethyl acetate extract had the highest total flavonoid content, with a value 63,138 mgQE/g. The antioxidant activity of quercetin was found to be 13600 FeEAC mol/g, while the potential antioxidant activity of the ethanol extract in the test sample was 2227,926 FeEAC mol/g. Further, the IC50 value for the ethyl acetate extract of Tetracera indica was 23,877 µg/mL. Based on the results, the Tetracera indica plant can be developed as an antioxidant and anti-cancer agent
