Abstract: Barringtonia asiatica L. Kurz (keben) seeds are one of the parts of medicinal plants that are used by the Tomini people as a medicine for eye infections. This study aims to determine the antibacterial activity of the keben seeds fractions by using the diffusion method (cylinder plate) and to identify antibacterial compounds from the fraction that has the highest antibacterial activity value using TLC-bioautography method and chromogenic reagents. The results of the antibacterial activity test showed that the n-hexane, ethyl acetate, and water fractions from keben seeds had an antibacterial activity with the highest inhibitory diameter in the ethyl acetate fraction, namely 21.08±0.72 (mm±SD) in Staphylococcus aureus and 21.53±1.00 (mm±SD) in Escherichia coli bacteria. The results of thin layer chromatography (TLC)-bioautography of the ethyl acetate fraction against Staphylococcus aureus suggest that compounds that have antibacterial activity are steroids (Rf 0.75 and 0.78) and terpenoid compounds (Rf 0.3) in Escherichia coli bacteria. Based on these results, the ethyl acetate fraction of keben seeds can be considered as a source of the antibacterial natural compounds.
