Abstract: | Warfarin is an oral anticoagulant drug used to treat thromboembolism-related diseases. Warfarin has a narrow therapeutic index and may interact with drugs and herbs thus requires careful monitoring of its pharmacological effects. Determination of blood warfarin levels is one of the methods of monitoring side effects due to drug interactions and consequently dose adjusments. This article is a literature review to provide an overview of the methods used to analyze the concentration of warfarin in blood plasma and its sample preparation technique. The method used in this article review is a search through the PubMed® database. Results showed that the methods commonly used for determination of warfarin in plasma are highperformance liquid chromatography (HPLC) and ultra-HPLC with ultraviolet (UV), fluorescence (FLD), diode array (DAD), and mass spectroscopy (MS) detectors. Other methods are UV/Vis or fluorescence spectrophotometry and electrophoresis. The HPLC with mass spectroscopy detector is widely used to analyze warfarin and the most preferred sample preparation method is protein precipitation using acetonitrile as a protein precipitant and the addition of acid to increase the recovery. |
