Abstract: Off-label drug use in obstetrics-gynaecology patients requires vigilance because of the risk for pregnancy. Although there is still debate, data on its use profile in clinical practice is still lacking. This study investigates off-label drugs in obstetric and gynaecological patients in private hospitals, especially Misoprostol. This study used a descriptive observational approach to hospital medical record data. Retrospective research data collection at RSU Bunda (RS X) for July 2017 - December 2017 and RS Sinar Kasih (RS Y) for the period January 2018 - December 2018. The number of samples was determined using the Slovin formula, with a sampling technique systematic random sampling at RS X and total sampling at Y Hospital. The total sample size was 220 RM at RS X 130 and RS Y 90. It was conducted between January 2019 and March 2019, and the results were analyzed univariately using several related references. The results showed that 26.92% of the off-label drug misoprostol was used at X Hospital and 4% at Y Hospital. The off-label drug found was Misoprostol which was included in the off-label indication category (Missed Abortion, Incomplete Abortion, Blighted ovum, and labour induction in Serotinus, PROM, and IUFD cases), off-label doses (50 µg, 100 µg and 400 µg), and off-label routes of administration (sublingual and vaginal).
